Poster text: Improving Blood Tests for Cancer Patients Mitigation of interference in blood compatibility testing caused by monoclonal antibody therapies using baitbodies Antibody therapies interfere with blood compatibility tests Baitbodies bind and neutralize the cancer therapy antibodies Baitbodies created from IgG Fc-region and CD38 / CD47 Epitope • Cancer immunotherapy uses monoclonal antibodies to target the tumor-associated "don't kill me"- antigens CD38 & CD47 • Anti-CD38 and Anti-CD47 antibodies trigger macrophage-mediated phagocytosis in Multiple Myeloma cells • Indirect antiglobulin test is a requirement for blood transfusions which Multiple Myeloma patients regularly need • Immunotherapy interferes with indirect antiglobulin test results, because both antigens are also expressed on donor Clumping Incompatible Blood Coombs Reagent Compatible Blood Doesn't Clump • The recombinant protein was synthesised in Expi293F cells • This fusion mimics the natural antigen structure where the FC-part stabilizes the protein and allows dimerization • The result is a soluble, well-folded molecule capable of specifically binding antibodies directed against CD38 or CD47 Baitbodies successful in 9 patient samples for 3 Treatment Reference Chart for Indirect Antiglobulin Test Because of high CD47 expression in red blood cells and high Magrolimab doses in patients the Magrolimab sample still agglutinated in this experiment, even with in-vitro binding affinity.
