Nicotine Conditioned place preference (CPP) protocol after sham (right) or MPEG4Ch (left) infusion. Usually, a low dose of nicotine wouldn't induce a preference in the CPP.
However, inhibiting the IPN with MPEG4Ch, the activation by nicotine in the IPN is reduced, while the response to low-dose nicotine in the VTA is enhanced. This is sufficient to induce a preference to low-dose nicotine in CPP.

We developed a newly tethered antagonist, named MPEG4Ch, to target the β4 subunit of the nicotinic acetylcholine receptors. MPEG4Ch treat-
ment of IPN brain slices markedly decreased the amplitude of
nicotine-induced currents in b4E61C mice.
