Schematic diagram showing SL biosynthetic pathways. CL, a common precursor of SLs, is synthesized from all-trans-β-carotene through sequential reactions catalyzed by D27, CCD7, and CCD8. The CYP711A family enzymes convert CL to CLA, and subsequent BC-ring formation of CLA leads to canonical SLs. These are classified into strigol-type and orobanchol-type SLs based on the stereochemistry of the C ring. Noncanonical SLs, characterized by an incomplete ABC-ring system, are believed to be synthesized from CL, CLA, or MeCLA without BC-ring formation. In cowpea, VuCYP711A converts CL to CLA (Wakabayashi et al. 2019), which is further oxidized to 18-oxo-CLA by VuCYP722C. VuSRF stereoselectively converts 18-oxo-CLA to orobanchol (M. Homma et al. 2024a), which is acetylated to orobanchyl acetate by VuOAT (M. Homma et al. 2024b). In addition, VuSRF-like, a paralog of VuSRF, converts 18-oxo-CLA to a novel noncanonical SL, orobanchonoic acid, via orobanchol. Updated model of hormonal crosstalk upon host-induced seed germination of weedy witchweeds and broomrapes.
