We applied ligation-free proximity extension and controlled amplification by ProPER to detect life cycle stages of RNAs directly by in situ imaging based on their proximity to life cycle markers (for eg. 18S rRNA or U1-snRNA). ProPER can also be multiplexed to detect multiple proximities even on the same molecular target and thanks to its modular and programmable design, it can supports in situ logic operations and multivalent proximity detection.
